摘  要:目的:探讨ApoA1-75G/A和+83C/T基因多态性与乳腺癌易感性之间的相关性。方法:研究对象为乳腺癌组217例,对照组221例。PCR-RFLP法检测ApoA1-75G/A和+83C/T基因多态性,采用SPSS13.0统计软件进行数据分析。结果:两组研究对象均存在ApoA1-75G/A和+83C/T基因多态性。ApoA1-75G/A多态性与乳腺癌发病,尤其是绝经前乳腺癌患者发病明显相关(OR=1.270,P=0.020;OR=1.445,P=0.003),与乳腺癌临床特征(肿瘤大小、淋巴结转移、病理分级以及雌孕激素受体状态)无明显相关。+83C/T多态性与乳腺癌发病明显相关(OR=1.517,P=0.005),与乳腺癌临床特征(月经状态、肿瘤大小、淋巴结转移、病理分级以及雌孕激素受体状态)无明显相关性。结论:ApoA1-75G/A和+83C/T基因多态性增加了乳腺癌发病的危险性,可能为乳腺癌发病的危险因素。OBJECTIVE: To evaluate the role of the functional ApoA1 polymorphisms (- 75 G/A and +83 C/T) as genetic markers for breast cancer susceptibility. METHODS: The polymerase chain reaction and restriction enzyme digestion (RFLP-PCR) were used to characterize the variations of the ApoA1 gene in 217 unrelated patients with breast carcinoma and 221 healthy control subjects. The data was analysed with SPSS 13.0 software. RESULTS:There were ApoA1 --75 G/ A and + 83 C/T polymorphisms in all subjects. A positive association was found between ApoA1 --75 G/A allele carriers and breast cancer risk (OR-= 1. 270, P = 0. 020). Regarding prognostic indicators, a sig nificant association was found between ApoA1 --75 G/A allele carriers and the premenopausal status of breast cancer patients (OR = 1. 445, P=0. 003). No association was found between ApoA1 --75 G/A allele carriers and breast cancer clinical feature including tumor size, lymph node metastasis, histological grade, estrogen and progesterone receptor. A positive association was found between ApoA1 +83 C/T allele carri- ers and breast cancer risk (OR=1.517,P=0.005). But no association was found between ApoA1 +83 C/T allele carriers and breast cancer clinical feature (including menopausal status, tumor size, lymph node metastasis, histological grade, estrogen and progesterone receptor). CONCLUSION: ApoA1 --75 G/A and +83 C/T polymorphisms can increase the pathogenetic risk of breast cancer, and may be the risk factors of breast cancer.
