卵白蛋白诱导的急性过敏性腹泻小鼠结肠组织的病理学特征

摘　　要：目的 建立急性过敏性腹泻模型,观察结肠组织的病理学特征.方法 5～6周龄雌性BALB/c小鼠随机分为实验组和对照组;用卵白蛋白（OVA）和完全弗氏佐剂对实验组小鼠隔1周2次腋窝皮下预致敏,第2次致敏1周后2组小鼠均每隔1d给予等量溶解在灭菌PBS的OVA灌胃;于第10次灌胃后处死小鼠,取血分离血清测OVA-IgE,取结肠近端组织行HE染色观察肠组织病理改变并在油镜下计数嗜酸性粒细胞.结果 与对照组相比,实验组小鼠结肠黏膜充血水肿,上皮排列不规则,腺区有大量炎细胞浸润,固有层可见大量嗜酸性粒细胞,血清OVA-IgE水平显著升高.结论 OVA 可诱导过敏性腹泻炎症,结肠黏膜组织以充血水肿和大量嗜酸性粒细胞浸润为特征.Objective To construct allergic： diarrhea model and observe pathological changes of colon. Methods The female BALB/c mice（ 5 -6 Week-old ） were randomly divided into control grnup and experimental group. The mice in experimental group were primed systemically twice with 1 mg of OVA in 100ul CFA suheutaneously in axilla eveu other week. One week after the second systemic priming,the mice in the two groups were delivered 5rag OVA twice and then 50rag of OVA dissolved in 350btl sterile PBS by intragastric administration every, other clay. Results Compared with control group,hyperemia and edema of mucosa and more inflammatory cells couhl be observed in proximal colon tissue,with significant eosinophils infiltrated in lamina propria,and there was a significant increase in OVA-IgE level in experimental group. Conclusion OVA can induce allergic diarrhea with characteristic of hyperemia and edenm of mucosa and eosinophils infihration in lamina propria.
