摘 要:以高蛋白、抗条纹叶枯病的红米水稻‘Kasalath’和本实验室培育的低直链淀粉含量、株型好、产量高的水稻‘上师大3号’为亲本进行杂交。利用蜡质基因(Waxy)在第1内含子5′端剪切位点上游55 bp处CT重复序列的重复数与直链淀粉含量呈负相关的特性,设计了1对SSR引物,利用与抗条纹叶枯病基因(Stv-bi)紧密连锁的SSR标记RM287设计了另1对SSR引物,结合田间表现,利用这两对引物对两亲本的杂交后代进行基因型选择,成功培育出具有‘上师大3号’蜡质基因和‘Kasalath’抗条纹叶枯病基因的红米水稻—‘上师大6号’。大田观察显示,‘上师大6号’水稻能够高抗条纹叶枯病,株高适中,穗型较密。经农业部稻米及制品质量监督及检验测试中心分析显示,‘上师大6号’水稻精米直链淀粉含量为12.0%,糙米蛋白质含量为13.5%。本研究开发的SSR标记是共显性标记,具有易于检测、重复性好、成本较低等优点,能够辅助选择具有低直链淀粉含量的Waxy的基因型及含抗条纹叶枯病基因的水稻。We use anti-Rice Stripe Virus,high-protein rice red rice 'Kasalath' and our laboratory bred with low amylose content,and good plant type,high yield rice 'Shangshida 3' as the parents for hybridization.Waxy gene(Waxy) in intron 1 of the 5′ splice site 55 bp upstream of the CT repeats at the repetition number was negatively correlated with amylose content of rice.Based on this characteristic,a pair of SSR primers P was designed;Based on the SSR marker RM287 sequence,which is closely linked to the stripe disease resistance gene(Stv-bi),another pair of SSR primers F was also designed.Combined with field performance,the two primer pairs were used to select,the offsprings.The red rice 'Shangshida 6',which content Waxy of 'Shangshida 3' and Stv-bi of 'Kasalath' was successfully cultivated.In the field,'Shangshida 6' rice displays high anti-rice stripe virus character,plant height is moderate,dense panicle.By the identification of Ministry of Agriculture and Testing Center for Quality Supervision,'Shangshida 6' have 12.0% amylose content of polished rice,13.5% protein content of brown rice.SSR markers developed in this study is co-dominant markers,which characteristics is easy-to-detection,good repeatability and low cost,can be used to assisted selection rice with low amylose content Waxy genotypes and Stv-bi.
