摘 要:利用表面活性剂-超声法提取野生刺五加果实中总黄酮,并用响应曲面法对提取条件进行优化。在单因素实验的基础上,根据Box-Behnken中心组合设计原理,选取超声波功率、液料比和表面活性剂十二烷基二甲基甜菜碱3个因素进行响应曲面分析,建立了野生刺五加总黄酮提取率的二次多项数学模型。得出其提取的最佳工艺条件为:超声功率110W,液料比96∶1(v∶m),十二烷基二甲基甜菜碱浓度为1.06%,提取时间为50min。在此条件下,刺五加中黄酮提取率达到8.72%。Surfactant-ultrasonic extraction was applied to gain flavonoids from the wild acanthopanax senticosus harms fruit. Response surface methodology(RSM)was applied to optimize the extraction conditions. Based on the single factor test,ultrasonic power,the ratio of liquid to solid and dodecyl dimethyl betaine(BS-12)concentrations were chosen as casual factors,and a three-factor and three-level Box-Behnken central composite design was performed to attain the highest extraction ratio of flavonoids. A second order quadratic equation for extraction of flavonoids was built. The optimal extraction conditions were determined as follows:ultrasonic power 110W,ratio of liquid to solid 96∶ 1(v∶ m),BS-12 concentration 1. 06%,and extraction time 50min. Under these conditions,the extraction ratio of acanthopanax senticosus flavonoids was 8. 72%.
