摘  要:目的观察辐射及多药耐药基因逆转剂反义寡核苷酸(mdr1ASON)联合5一氟尿嘧啶磁性白蛋白微球(5一Fu-MAMS)靶向治疗耐药裸鼠人胰腺癌模型的效果。方法构建人胰腺癌耐药细胞株(SW1990/Fu),建立裸鼠人胰腺癌耐药模型,选择适当的磁场,施加于肿瘤表面,瘤内注射mdr1ASON及5一Fu.MAMS,观察肿瘤生长,检测其对耐药裸鼠人胰腺癌的治疗效果。结果在外加磁场的磁导向作用下,5-Fu-MAMS能定位于肿瘤组织,联合mdr1ASON能显著抑制肿瘤生长,肿瘤体积抑制率达到85.00%,肿瘤重量抑制率达到87.74%,与其他组比较差异有统计学意义(P〈0.01)。结论辐射促转染的mdr1ASON联合磁性载药微球对肿瘤细胞具有较好的耐药逆转作用。Objective To investigate the reversal the muhidrug resisitanee (MDR) by MDR gene mdrl antisense oligodeoxynueletdes (ASON) and 5-fluorouraeil magnetie albumin miserospheres (5-Fu- MAMS) magnetic targeted therapy the human pancreatic cancer in nude mouse model. Methods To construct SW1990/Fu and nude mice experimental model, to investigate the targeted therapy effects of mdrlASON and 5-Fu-MAMS. Results 5-Fu-MAMS magnetic response was good, its could be well localized in the tumor tissue and markedly inhibit the growth of tumor. The inhibitory rate of tumor size was 85.00%, and that of tumor weight 87.74%. which can effectively degrade proliferating index and accelerate cell death of tumor cell. There were significant differences as compared with other groups(P 〈 0. 01 ). Conclusion Mdrl ASON and 5-Fu-MAMS had outstanding reverse effect drug fast on human pancreatic carcinoma model in nude mice.
