摘 要:[目的]对耐温酵母用于玉米秸秆同步糖化的发酵工艺进行考察。[方法]研究固液比、初始葡萄糖浓度和酶加入量对纤维素酶水解的影响,并对温度、固液比对中性蒸汽爆破预处理玉米秸秆(PCS)同步糖化发酵的影响和耐温酵母FE-B的同步糖化发酵工艺进行考察。[结果]水解体系中水解速率在一定范围内与酶加入量成正比,但随着酶加入量的增加,酶量对水解速率的贡献逐渐降低;酶解体系中的葡萄糖浓度对纤维素酶的水解速率具有很强的抑制作用,葡萄糖浓度越高,抑制越强;固液比在10%~30%,固含量越高,最终葡萄糖产率越高,且不影响纤维素水解率。利用基于耐温酵母FE-B的同步糖化发酵工艺水解和发酵120 h,反应体系中的乙醇浓度可达6.21%(W/W),纤维素水解率为88.70%。[结论]该研究可为开发木质纤维素生产燃料乙醇提供理论依据。[ Objedtive ] The study aimed to investigate the technology of the simultaneous sacchariiication and termentation (SSF) by using heat-resistant yeast. [ Method] The influence factors including the solid-liquid ratio, initial glucose concentration and cellulose adding amount on the hydrolysis with the cellulase were researched and the influences of the temperature and solid-liquid ratio on the SSF of the corn straw pretreated by the neutral steam explosion and the technology of the SSF by using heat-resistant yeast FE-B were investigated. [ Result] The hy- drolysis rate in the hydrolysis system was positive to the cellulase adding amount in a certain range, but with the increment of the cellulase adding amount, the contribution rate of cellulose dosage to the hydrolysis rate was decreased gradually. The glucose concentration in the hy- drolysis system had a very strong inhibition on the hydrolysis rate of the cellulase and the higher the glucose concentration, the smmger the inhi- bition. When the solid-liquid ratio was 10% - 30%, the higher the solid content, the higher the final glucose yield, which didn' l affect the hy- drolysis rate of cellulose. When the technology of the SSF by the heat-resistant yeast FE-B was used to hydrolyze and ferment tbr 120 h, the ethanol concentration in the reaction system was 6.21% (W/W) and the hydrolysis rate of cellulose was 88.70%. [ Conclusion~[ The research orovided a theoretical basis for develoDin~ the woodiness cellulose to produce the ethanol.
