摘 要:目的探讨维生素C和牛磺酸对铁过载小鼠心脏损伤的保护作用。方法采用腹腔注射右旋葡萄糖铁剂的方法,获得急性铁过载小鼠模型。急性铁过载小鼠再随机分组,分别给予维生素C(150mg.kg-1)、牛磺酸(420mg.kg-1)、1/2维生素C(75mg.kg-1)+1/2牛磺酸(210mg.kg-1)和0.5mL生理盐水灌胃;对照组给予0.5mL生理盐水灌胃,4周后,提取各组小鼠血浆测定血浆CK、LDH值,提取各组小鼠心脏测定心肌的MDA、GSH-PX、SOD值。结果维生素C组、牛磺酸组和1/2维生素C+1/2牛磺酸组小鼠的血浆CK活性显著低于铁过载组(P<0.05),但明显高于对照组(P<0.05);维生素C和牛磺酸对小鼠血浆CK活性的影响有协同作用(P<0.05)。铁过载组小鼠血浆LDH活性比其他4组小鼠血浆LDH活性明显升高(P<0.05),其他4组之间小鼠血浆LDH活性差异无统计学意义(P>0.05)。维生素C组、牛磺酸组和1/2维生素C+1/2牛磺酸组小鼠心肌MDA含量比铁过载组明显降低(P<0.05),但和对照组小鼠心肌MDA含量相比较差异无统计学意义(P<0.05)。维生素C和牛磺酸对降低小鼠心肌MDA的含量有协同作用(P<0.05)。铁过载组小鼠心肌SOD活性比牛磺酸组、1/2维生素C+1/2牛磺酸组和对照组小鼠明显降低(P<0.05),牛磺酸组、1/2维生素C+1/2牛磺酸和对照组心肌SOD活性3组之间无差异(P>0.05)。结论维生素C对铁过载引起的小鼠心脏损伤有保护作用;维生素C和牛磺酸在改善铁过载对小鼠所造成的心脏损伤方面有协同作用。Objective To investigate the protective effects of vitamin C and taurine on iron overload cardiomyopathy in mice. Methods A mouse model of myocardial iron overload was made by intraperitoneal injection of dextroglucose ehalybeate. Iron-overloaded mice were intragastricly administrated with 150 mg·kg^-1 vitamin C (Asc group), 420 mg·kg^-1 taurine (Tau group),75 mg·kg^-1 vitamin C plus 210 mg·kg^-1 taurine (Asc+Tau group) or 0.5 mL normal saline (iron overload group). Control mice received 0.5 mL normal saline only. The plasma creatine kinase (CK) and lactate dehydrogenase (LDH) activities were detected and myocardial malondialdehye (MDA) content and superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) activities were measured in all mice. Results Plasma CK activities and myocardial MDA concentrations in Asc group, Tau group and Asc+Tau group significantly decreased compared with iron overload group(P〈0.05),but obviously increased compared with control group (P〈0.05). Moreover,vitamin C and taurine had a synergistic effect (P〈0.05). Plasma LDH activities in iron overload group were significantly higher than those in other groups(P〈0. 05), but no differences were observed among other four groups(P〈0.05). Compared with iron overload group, plasma SOD activities significantly increased in Tau group, Asc +Tau group and control group (P〈 0.05), but there were no differences among the three groups (P〉0.05). Conclusion Vitamin C has a protective effect on myocardial damage induced by iron overload. In addition, vitamin C can synergize with taurine to improve iron overload cardiomyopathy in mice.
