摘 要:土壤中蕴含丰富的微生物资源,脂肪酶在微生物中分布广泛.为降低成本和扩大脂肪酶的应用,从富油土壤中自主筛选获得两株产脂肪酶菌株(分别命名为TS1和TS2),利用平板分离培养基进行纯化培养,对菌株的形态特征和生物学特性进行了研究.碱滴定法测定两株菌的产脂肪酶活性,在限定4 d的培养周期内考察温度和时间因素对菌株产酶活性的影响.结果表明,TS1在34℃条件下培养3 d可获较高酶活(2.40 U/mL);TS2的相对适宜温度范围较窄,在32℃条件下培养3 d酶活可达2.05 U/mL.Lipase widely exists in microorganism resources which are ample in soils.For cutting cost and broadening the application of lipase,two lipase-producing strains were isolated from oil-rich soil and named as TS1 and TS2,respectively.After being purified from flat separation culture mediums,the morphological and biological characteristics of the strains were studied.Their enzymatic activities were determined by titration.The effects of significant factors(temperature,culture time) on the enzymatic activities were investigated.The restricted culture time was 4 d.The results indicated that TS1 got its better enzymatic activity of 2.40 U/mL on the 3rd culturing day at 34 ℃ and TS2 got its better enzymatic activity of 2.05 U/mL on the 3rd culturing day at 32 ℃.
