摘  要:目的 研究家蝇胚胎细胞MDEC-07114抗菌肽对黑色素瘤细胞A375的抑制作用.方法 利用脂多糖诱导家蝇胚胎细胞MDEC-07114提取抗菌肽.将家蝇胚胎细胞抗菌肽分为40,80,160,320和640 μg/ml五个实验组,同时以PBS和培养基作为正常对照组.采用MTT比色法测定不同浓度抗菌肽作用下细胞生长曲线,流式细胞术观察抗菌肽对黑色素瘤细胞A375周期及凋亡的影响.结果 ①在不同浓度家蝇胚胎细胞抗菌肽作用下,黑色素瘤细胞A375的生长受到明显的抑制,药物浓度越高、作用时间越长,对细胞的抑制作用越强.②各浓度组抗菌肽引起A375细胞的G2/M期升高,高浓度组(>160 μg/ml)可引起G0/G1期降低,PI明显升高,与正常对照组相比差异具有统计学显著性意义(F=214.89,P<0.05).与正常对照组相比,各实验组引起的A375细胞凋亡率差异无统计学显著性意义(F=14.17,P>0.05).结论 家蝇胚胎细胞抗菌肽对肿瘤细胞的抑制作用随着药物剂量的增加和作用时间的延长而增强.家蝇胚胎细胞抗菌肽对A375细胞的抑制机理可能主要是阻滞细胞周期的正常发育.Objective To research the inhibitory action from the theembryonic cells MDEC-07114 of the musca domestica antibacterial peptide on the melanoma cells A375. Methods The logarithmic growth phase's embryonic cells of the musca domestica were induced by LPS. The antibacterial peptide was extracted from supernatant fluid. The antibacterial peptide was prepared 40,80,160,320 and 640 μg/ml five density groups by PBS,PBS and culture medium were been normal control group. The growth curves of A375 ceils in the above five density groups were maden by MTT method. The cell cycle and the cell apoptosis ratio of the A375 cells were detected in five density groups of antibacterial peptide by flow cytometry. Results Under effects of the different density antibacterical peptides,growths of A375 cells were inhibited obviously, and cell mortality rates rised along with time extension and medicine density increase. For A375 cells, five density antibactericaI peptides produced all rise of the G2/M,in the highly concentration groups (〉160 μg/ml) the G0/G1 reduced and PI advanced obviously,the apoptosis ratio had significant difference( F= 214.89, P〈0.05), compared with the normal control group. Compared with the normal control group,the apoptosisratio of the A375 cells in various density groups had not significant difference (F= 14. 17, P〉0.05). Conclusion The inhibitory activity of the antibacterical peptide from the embryonic cells of the musca domestica on tumour cells enhances with the time-extention and dose-increase. The inhibition mechanism of the antibacterical peptide from the embryonic cells of the musca domestica on A375 cells maybe arrest normal development of the cell cycle.
