摘 要:为了除去三叶委陵菜中总黄酮的杂质,选择以大孔吸附树脂对三叶委陵菜黄酮的吸附率、解析率为指标,考察了六种大孔吸附树脂对三叶委陵菜中总黄酮的分离纯化性能,筛选出了性能最佳的大孔吸附树脂,同时采用高效液相色谱法进行分析检测进而表征了最佳型号的大孔吸附树脂对三叶委陵菜中总黄酮分离纯化的效果;分析了原液浓度、pH和树脂用量以及洗脱剂的浓度和pH对吸附解析效果的影响。实验结果表明,大孔吸附树脂D101对三叶委陵菜总黄酮有很好的吸附和解析性能,并确定了最佳的吸附脱附条件为:吸附液浓度为2.0 mg/mL、pH=6、树脂用量与吸附液量比(g/mL)为1∶30;平衡浓度达到0.36 mg/mL时,即树脂已经达到吸附饱和,饱和吸附量为49.1 mg/g干树脂;洗脱剂乙醇浓度为50%,pH=6。In order to purify the total flavone of Potentilla freyniana,the separation and purification abilities of 6 kinds of macroporous adsorption resins for the flavones of Potentilla freyniana were studied.The adsorption and desorption were used as the indexes.The most available resin was selected and the purification effect was estimated by HPLC analysis.The influences of the concentration and pH of solvent on adsorption and desorption and resin quantity were studied.The results showed that D101 macroporous resin had the best adsorption and desorption performance.The optimal adsorption and desorption conditions were: adsorption liquid concentration 2.0 mg/mL,pH=6,the ratio of resin to adsorption fluid volume 1∶ 30(g/mL).When the equilibrium concentration was 0.36 mg/mL,the resin reached saturation adsorption.The saturation adsorption was 49.1 mg/g(dry resins).Ethanol of 50%(pH=6) was used as desorption solvent.
