摘  要:目的:阐明胃癌生长对氨基酸代谢的影响以及对某些氨基酸的特殊需求。 方法:采用RP-HPLC技术检测12例胃癌病人肿瘤组织20 种游离氨基酸含量,且与肿瘤体积进行相关性和多元逐步回归分析。 结果:胃癌组织游离谷氨酰胺、精氨酸、蛋氨酸含量较正常胃粘膜明显增高(P< 0.05),而丙氨酸、色氨酸含量较正常胃粘膜明显降低(P< 0.05)。同时胃癌组织游离谷氨酰胺、精氨酸、蛋氨酸含量与肿瘤体积呈正相关,而丙氨酸与肿瘤体积呈负相关。 结论:随着胃癌生长,肿瘤细胞特别需求谷氨酰胺、蛋氨酸、精氨酸作为蛋白质、核酸合成的原料。Objectives:In order to illuminate the effect of gastric carcinoma growth on amino acid metabolism,and special requirement of tumor for some amino acids. Methods:Levels of 20 free amino acids in tumor tissue were measured from 12 patients with gastric carcinoma and normal stomach mucosa as controls using reversed phase high performance liquid chromatography.Correlation was further studied between tumor volume and 20 free amino acids in tumor tissue. Results:Content of free glutamine,methionine and arginine in gastric carcinoma tissue were significantly increased when compared with those of normal stomach mucosa( P <0.05).On the other hand,content of tumor tissue free alanine and tryptophan were significantly decreased when compared with those of normal stomach mucosa( P <0.05).A positive correlation was found between tumor volume and tumor tissue free glutamine,methionine or arginine( r =0.923,0.932,0.888, P <0.01),and a negative correlation was found between tumor tissue free alanine and tumor volume( r =-0.711, P <0.01). Conclusions:Tumor requires a large amount of amino acids in growth,particularly for glutamine,methionine,arginine as substrate of tumor protein and nucleic acid systhesis,resulting in change of amino acids metabolism.
