摘 要:背景与目的:恶性黑色素瘤具有放射敏感异质性,属于增殖速度缓慢的肿瘤,如何提高其放射治疗敏感性和增益比成为恶性黑色素瘤放射治疗的瓶颈。本研究主要探讨金纳米棒对人恶性黑色素瘤A375细胞放射敏感性的影响。方法:利用MTT法检测金纳米棒的细胞毒性;透射电子显微镜观察金纳米棒进入细胞的情况;采用6 MeV电子线照射金纳米棒温育后的A375细胞,利用流式细胞仪检测细胞的凋亡和周期情况;克隆形成实验检测细胞的放射敏感性改变。结果:金纳米棒在50μg/mL浓度范围内对A375细胞生物学活性无明显影响;透射电镜下观察到金纳米棒通过内吞作用进入细胞质内;经金纳米棒温育后细胞G_2/M期比例显著提高(P<0.05),在联合高能电子线照射后恶性黑色素瘤A375细胞的凋亡率和放射敏感性也显著提高,DMF=1.34(P<0.05)。结论:金纳米棒可有效提高恶性黑色素瘤A375细胞对高能电子束的放射敏感性。Background and purpose:The radiation sensitivity of malignant melanoma has heterogeneity, with slow proliferation.So how to improve the radiosensitivity and therapeutic gain factor is the bottle neck of the radiation therapy of malignant melanoma.Our study mainly discussed the effect of gold nanorods on human malignant melanoma cell radiosensitivity.Methods:The cytotoxicity of gold nanorods was assessed by MTT assay. The transmission electron microscope was used to detect the gold nanorods in cells.6 MeV clinical electron beams irradiated the melanoma A375 cells after incubating with gold nanorods.The cell cycle and apoptosis were examined by flow cytometry(FCM);the radiosensitizing effect was performed by clonogenic assay.Results:Gold nanorods in the 50μg/mL concentration range were safe to the biological activity of A375 cells and the gold nanorods can enter the cytoplasm by endocytosis.After incubating with gold nanorods,the melanoma A375 cells significantly accumulated in the G_2/M phase(P0.05).The radiosensitivity of melanoma A375 cells and the cell apoptosis rate were increased significantly in gold nanorods group combined irradiation group,DMF=1.34(P0.05).Conclusion:Gold nanorods can effectively improve the radiation sensitivity of malignant melanoma A375 cells to high-energy electron beams.
