摘  要:目的对转人乳铁蛋白基因大米(hLF大米)与亲本大米(PR大米)主要营养素在体内的消化代谢及蛋白质的营养价值进行初步比较评价。方法以6头近交系五指山小型去势公猪为实验动物,进行回肠末端造瘘手术安置T型瘘管,采用交叉法分别饲喂hLF大米、PR大米,收集回肠末端消化产物分析比较两种大米主要营养素的体内消化率,采用经消化率校正的氨基酸评分法(PDCAAS)对两种大米蛋白质的营养价值进行初步评价。结果 hLF大米与PR大米在蛋白质/氨基酸、碳水化合物、脂肪、纤维素的消化率上均无显著差异。人乳铁蛋白基因的插入使hLF大米中各氨基酸的含量配比得到了一定的优化,且其PDCAAS比亲本大米提高了2.66%。结论 hLF大米与PR大米在五指山小型猪体内主要营养素的消化代谢上满足"实质等同性"的要求,PDCAAS的结果显示hLF大米中蛋白质的质量略有提高。Objective To study the digestibility of main nutrients in genetically modified rice expressing human lactoferrin(hLF rice) and to assess its protein quality compared with the parental rice(PR rice).Method Six Wuzhishan mini-pigs inbred-line were used as the experimental animals.T-cannulas were equipped at their terminal ileums by surgical operation.After that,the pigs were randomly divided into two groups,and fed with hLF rice and PR rice by crossover method.Ileal digesta was collected for analysis of main nutrients’ digestibility.Compared with PR rice,the nutritional quality of protein in hLF rice was also assessed by protein digestibility corrected amino acid scores(PDCAAS) method.Results The hLF rice was not statistically different from its parental rice in terms of protein/amino acids,carbohydrate,fat and fiber digestion and absorption in Wuzhishan mini-pigs.The ratio of amino acids content in hLF rice was optimized as result of exogenous hLF gene insertion,and the PDCAAS of hLF rice was higher than that of PR rice by 2.66%.Conclusion The digestibility of main nutrients in hLF rice and PR rice are substantially equivalent.The nutritional quality of protein in hLF rice is slightly superior to PR rice according to PDCAAS.
