摘 要:目的探究海带多糖(laminar polysaccharide,LP)对肾上腺素代谢产物H2O2、甲醛和甲胺损伤血管内皮依赖性舒缩功能的影响。方法制作大鼠离体胸主动脉环,将血管环分为对照组(加入等量的K-H液)、模型组(分别给予H2O2、甲醛和甲胺孵育)、海带多糖高剂量组(LP-H,给予H2O2、甲醛和甲胺孵育的同时加入高剂量LP)和海带多糖低剂量组(LP-L,给予H2O2、甲醛和甲胺孵育的同时加入低剂量LP),孵育后分别测定PE预刺激后血管环对乙酰胆碱(acetylcho-line,ACh)的舒张反应,及血管环对去甲肾上腺素(norepi-nephrine,NE)的收缩反应。结果 H2O2孵育后,对照组、LP-H、LP-L的舒张反应明显高于模型组(P<0.05);但各组血管环的收缩反应差异无显著性。甲醛孵育后,对照组、LP-H的舒张反应明显低于模型组(P<0.05);对照组、LP-L和LP-H的血管收缩反应低于模型组(P<0.05)。甲胺孵育后,对照组和LP-H的血管舒张反应明显高于模型组(P<0.05);对照组、LP-H的血管收缩反应低于模型组(P<0.05)。结论海带多糖对肾上腺素代谢产物H2O2、甲醛和甲胺所致的血管内皮依赖性舒缩功能的损伤具有一定的保护作用。Aim To explore the effect of laminar polysaccharide on the vascular endothelium-dependent relaxation and contraction injured by the metabolic products of adrenaline.Methods The rat′s aortic rings in vitro were set up,and then the aortic rings were divided into control group,model group,LP-H and LP-L.H2O2,formaldehyde and methylamine were added to the model,LP-H and LP-L groups respectively.High dose of LP was added to LP-H group while low dose to LP-L group at the same time.Control group was added KH only.The tensions of vascular relaxation induced by ACh after stimulation of PE and the tensions of contraction induced by NE were recorded respectively.Results After H2O2 was added,the relaxations of LP-H,LP-L,control group were more obvious than those of model group(P0.05) and there was no difference in the contractions.After formaldehyde was added,the relaxations of model group were more obvious than those of control group and LP-H.And the contractions of LP-H,LP-L,control group were less obvious than those of model group(P0.05).After methylamine was added,the relaxations were more obvious than those of model group and the contractions were less obvious than those of model group in LP-H and control group.Conclusion Vascular endothelium-dependent relaxation and contraction can be damaged by the metabolic products of adrenaline,and laminar polysaccharide can partially protect these damages.
