摘  要:以提高酱油曲霉ATCC20235产巯基氧化酶(SOX)产量为目的,选择碳源、氮源、发酵培养基初始pH及8种金属离子分别进行单因素实验,在此基础上选取8个主要因素进行正交实验。结果表明,以碳源(蔗糖)30g/L、氮源(鱼粉蛋白胨)6.8 g/L,K2HPO4·3H2O 15 g/L,MnCl2·4H2O 2 mg/L,BaCl2 500μg/L,H3BO3 1 mg/L,起始pH 8.5的优化培养基在30℃下,220 r/min培养72 h,获得发酵液巯基氧化酶产量达240U/L,优化后的酶产量比在专利文献中记载的发酵培养基及培养条件下的产酶量提高了10倍。In order to enhance the yield of SOX produced by Aspergillus sojae ATCC20235 ,the influence of car- bon source, nitrogen source, initial pH and other 8 minerals were first investigated for the first time using the single factor experiment. The orthogonal designs L1s (37 × 21 ) was carried out based on the result of the single factor experi- ment, and the optimal condition were listed as follow : carbon sources 30g/L, nitrogen sources 6.8g/L, K2 HPO4· 3 H2O 15g/L,MnCl2·4H2O 2 mg/L,BaCl2 500μg/L,H3BO3 1 mg/L, and pH 8. 5. Using this optimized media, the yield of SOX can achieve 240U/L. that is 10 times of the initial quantity when using the culture medium given by the US patent.
