脂质体介导的融合基因pCEAcd/tk转染肺癌细胞株效率的差异性

摘　　要：对脂质体介导的体外肺癌细胞DAN转效率的差异性进行研究。方法 :提取质粒 pCEAcd/tk ,脂质体包裹后转染 6种肺癌细胞株 ,G418筛选 ,计数细胞阳性克隆数。结果 :6种肺癌细胞株中 ,导入了pCEAcdt/tk并获得稳定表达的细胞阳性克隆数差异较大 ;脂质体ESCORT -DNA混合物与细胞孵育时间不同对转染效率也有一定的影响。结论Objective: To investigate the variation in transfection efficiency of cationic lipid-mediated pCEAcd/tk among different lung cancer cell strains in vitro. Methods: pCEAcd/tk was extracted and transferred to differental lung cancer cell strains by cationic lipid-mediated method. It was selected by G418 and the number of positive clones was calculated under microscope. Results: There are different positive clones among 6 lung cancer cell strains. The transfection efficiency was influenced by different incubate time between ESCORT-DNA mixture and lung cancer cells. Conclusions: There was a considerable variation in transfection efficiency of cationic lipid-mediated gene transfer among different cell strains.
