摘 要:目的 研究转化生长因子-β1(TGF-β1)在大鼠腹膜间皮细胞(RPMCs)转分化中的作用及机制.方法 体外培养SD大鼠原代腹膜间皮细胞,静止24h后,随机分为正常对照组(不加TGF-β1处理)及TGF-β1(10ng/ml)刺激组.采用RT-PCR法及Western blotting检测TGF-β1处理后不同时点NADPH氧化酶亚单位p67phox、α-平滑肌肌动蛋白(α-SMA)、E-钙粘素(E-cadherin)及Ⅰ型胶原(CollagenⅠ)mRNA和蛋白的表达情况.结果 TGF-β1刺激RPMCs能导致E-cadherin mRNA和蛋白表达下调,p67phox、α-SMA、CollagenⅠmRNA和蛋白表达上调,呈现一定的时间依赖性.结论 TGF-β1诱导了大鼠腹膜间皮细胞转分化,NADPH氧化酶可能在其转分化过程中发挥重要作用.Objective To investigate the effects of TGF-β 1 on EpitheliaI-Mesenchymal Transition of rat peritoneal mesothelial cells (RPMCs) Methods After synchronization for 24 h, primarily cultured rat RPMCs were randomly assigned to 2 groups: group A (control), group B (TGF- β, 1, 10ng/ml). Growth arrested and synchronized RPMCs were stimulated by 10ng/ml TGF- β 1 for different times. The mRNA and protein expression levels of NADPH oxidase subunit p67phox, E-cadherin, α -SMA and Collagen I were measured by RT-PCR and Western blotting, respectively. Results TGF- β 1 up-regulated mRNA and protein expression of NADPH oxidase subunit p67phox, α-SMA and Collagen I , and down-regulate mRNA and protein expression of E-cadherin in rat RPMCs. Conclusion RPMCs can be transdifferentiated into myofibroblast with TGF- β1, which may be related to the activation of NADPH oxidase.
