氟他胺的合成工艺改进

摘　　要：报道了以３三氟甲基苯胺为起始原料进行氟他胺合成的工艺改进方法，即，碳酸氢钠作为吸氯化氢剂的条件下，２甲基丙酰氯（ Ⅱ） 与３三氟甲基苯胺（ Ⅰ） 反应得到２甲基Ｎ〔（３三氟甲基） 苯基〕丙酰胺（ Ⅲ） ，再用发烟硝酸，硫酸，在－ ２ ℃下进行混酸硝化得到相应的２甲基Ｎ〔４硝基３（ 三氟甲基） 苯基〕丙酰胺（ 氟他胺Ⅳ） ．An improved synthesis of flutamide using 3 trifluoromethy aniline as the starting material was described.Treatment of the 3 trifluoromethyl aniline(Ⅰ) with 2 methylpropinonyl chloride(Ⅱ)in the presence of an anhydrous sodium bicarbonate as the hydrogen chloride acceptor gave the 2 methyl N 〔3 trifluoromethyl)phenyl〕propionamide(Ⅲ),and then it was nitrated at -2℃ with a fuming nitro sulfuric acid mixture to yield the corresponding 2 methyl N 〔(4 nitro 3 trifluoromethyl)phenyl〕propanamide(flutamide,Ⅳ).
