摘 要:目的:采用体外培养细胞基因突变试验,评价乙嘧酚磺酸酯原药对中国仓鼠卵巢CHO细胞基因HPRT立点和小鼠淋巴瘤L5178Y细胞基因TK位点的诱变性。方法:使用乙嘧酚磺酸酯原药对CHO细胞和L5178Y细胞进行染毒,实验设置5、15、50、150μg/ml共4个浓度组,分别对CHO细胞进行集落形成率,HPRT位点突变频率的测定;对L5 178Y细胞进行接种效率,相对存活率,TK位点突变频率的测定。结果:随着乙嘧酚磺酸酯原药剂量的逐渐增高,CHO细胞的集落形成率较阴性对照组明显降低(P<0.01);L5178Y田胞的平板接种效率和相对存活率与阴性对照组相比均明显下降,但是设计的4个剂量组基因突变率与阴性对照组比较差异均无统计学意义(P>0.05)。结论:在本实验条件下,乙嘧酚磺酸酯原药对CHO细胞和L5178Y细胞具有明显的细胞毒性,但其对细胞基因HPRT应点和TK位点的突变率无明显影响。OBJECTIVE:To investigate the effects of bupirimate on HPRT gene of ovarian cells and the TK gene of lymphoma cells of mouse in vitro culture system.METHODS:CHO cells and L5178Y cells were treated with bupirimate at different concentrations(5,15,50,150μg/ml).Determination of cytotoxicity,cell inoculation efficiency,relative survival rate and mutation frequency were performed.RESULTS:The relative survival rate and suspension growth rate of CHO cells and L5178Y cells decreased significantly with the increasing doses of bupirimate.But the mutation frequencies of HPRT and TK genes induced by bupirimate(5-150μg/ml) were not higher than that of spontaneous mutation frequency of L5178Y cells.CONCLUSION:Bupirimate did not show obvious mutagenic effects on HPRT gene in CHO cells and TK gene in L5178Y cells.
