摘　　要：目的探索Genistein对恶性黑素瘤LiBr细胞Livin基因表达的影响。方法分别在基因和蛋白水平上应用RT-PCR和Western blot检测、确认恶性黑素瘤LiBr细胞Livin的表达;应用RT-PCR检测Genistein作用LiBr细胞后Livin的表达变化,并初步探讨其剂量及时间效应。结果经RT-PCR和Western blot检测,恶性黑素瘤LiBr细胞高表达Livin基因;Genistein作用LiBr细胞后能显著下调Livin基因表达,并且下调过程呈剂量和时间依赖关系,40μmol/L可达到最大沉默效应,其下调Livin mRNA的最大效应出现在作用后96h。结论Genistein能显著下调恶性黑素瘤细胞Livin基因表达。Objective To investigate the effects of Genistein on expression of livin in human malignant melanoma LiBr cells.Methods Both at Gene and protein level,RT-PCR and Western blot are respectively used to confirm the delivery of Livin gene in malignant melanoma LiBr cells.After Genistein is used,we analyze the change in the delivery level of Livin gene as well as dosage and time through RT-PCR.Results Genistein could effectively knock down the Livin expression at mRNA in dose and time dependent manners,40 μmol/L of which achieved the highest knockdown effect on mRNA at 72 h after using Genistein.Conclusion Expression of livin in LiBr cells could be efficiently knocked down by using Genistein.
