摘  要:为获得高产碱性β-葡萄糖苷酶的产纤维素酶菌株,利用碱性羧甲基纤维素钠平板筛选和β-葡萄糖苷酶平板筛选法相结合,从植物腐败堆积土壤中分离到1株高产碱性β-葡萄糖苷酶的产纤维素酶克雷伯氏杆菌,并对其进行初步酶学鉴定。结果表明:该菌胞外分泌液具有内切葡聚糖酶、外切葡聚糖酶及β-葡萄糖苷酶的活性,其中以β-葡萄糖苷酶活性最高,为48.9U/mL。其所产纤维素酶的最适反应pH值为10.0,最适反应温度为35℃,且Ca2+、Mg2+、Zn2+和Mn2+等金属离子对该酶具有较强的抑制作用。In order to get a cellulase-high-yielding bacterium,CMCNa plate and β-glucosidase plate screening method were used to screen a bacteria strain,and its enzymology properties were preliminarily identified.The results showed that a facultative anaerobic strain which produced cellulase was obtained from the CMCNa plate and β-glucosidase plate screening.This strain was identified as Klebsiella sp.,and termed as strain CBG-08.The secretion of this strain showed activities of endoglucanase,cellubiohydrolase and β-glucosidase,β-glucosidase had the highest activity.The enzyme had the highest activity when pH was 10.0 and temperature was 35℃,and metal elements of Ca2+,Mg2+,Zn2+ and Mn2+ could inhibit enzyme activity.
