摘 要:探讨3种菌株所产木聚糖酶组分及其酶解产物的差异,为酶制剂和功能低聚木糖的研发和应用提供理论依据。采用SDS-PAGE电泳,研究黑曲霉C71、康宁木霉M46及XYNB重组工程菌G81所产的木聚糖酶同工酶。结果表明,C71、M46及G81分别产3种、4种和2种木聚糖酶同工酶;以桦木木聚糖为底物,在酶添加量为70U/g、底物20g/L、反应时间10h条件下进行酶解,采用TLC及HPLC分析酶解产物。TLC结果表明,C71和M46酶解液中含有木糖、木二糖和木三糖;G81酶解液中含有木二糖、木三糖和木四糖。HPLC结果表明,C71酶解液中木糖、木二糖和木三糖的质量浓度分别为2.4、1.3和1.7g/L;M46酶解液中木糖、木二糖和木三糖的质量浓度分别为1.5、1.8和2.1g/L;G81酶解液中木二糖、木三糖和木四糖的质量浓度分别为2.6、2.7和1.4g/L。不同菌株所产木聚糖酶组分及其酶解产物存在差异,低聚木糖的研发应根据酶学特性,选择不同的单一或复合酶制剂。Xylanases were produced by Aspergillus niger C71,Trichoderma koningii M46 and recombinant XYNB C81.These xylanase isozymes were analyzed with SDS-PAGE electrophoresis.The hydrolyzing conditions of xylanases were as follow: substrate is birchwood,dosage of enzyme is 70 U/g,substrate concentration is 20 g/L,the hydrolysis time is 10 h.The hydrolysates of xylanases from three different strains were detected by using TLC and HPLC.The results showed that the components of xylanase were N-1,N-2,N-3(produced by Aspergillus niger G71);Y-1,Y-2,Y-3,Y-4(produced by Trichoderma koningii M46) and Z-1,Z-2(produced by the recombinant XYNB G81).TLC and HPLC analysis indicated that the hydrolysates of xylanases produced by A.niger C71 and Trichoderma reesi M46 contained xylose(2.4 g/L,1.5 g/L,respectively),xylobiose(1.3 g/L,1.8 g/L,respectively) and xylotriose(1.7 g/L,2.1 g/L,respectively).The hydrolysates of xylanase produced by XYNB G81 contained xylobiose(2.6 g/L),xylotriose(2.7 g/L) and xylotetraose(1.4 g/L).The experimental results provided theoretical basis for the development of XOS.
