摘  要:目的检测不同恶性程度胶质瘤组织NAD(P)H氧化酶(NOX)的表达及其产物R0s的含量,探讨NOx及细胞内ROS与胶质瘤增殖及恶性程度的关系。方法收集南方医科大学珠江医院神经外科自2007年8月至2010年8月手术切除的人脑胶质瘤标本30例.其中低级别胶质瘤(Ⅰ和Ⅱ级)10例、Ⅲ级胶质瘤10例和Ⅳ级胶质瘤10例,另取10例行内减压术的脑损伤患者的正常脑组织作为对照组。RT-PCR检测4组脑组织NOX1~5mRNA的表达,流式细胞仪测定脑组织ROS的含量,Western blotting和免疫荧光染色分别检测脑组织NOX蛋白的表达和分布。结果4组脑标本NOX1~5mRNA、ROS含量均不同,差异均有统计学意义(P〈0.051,且NOX4 mRNA、ROS含量在正常对照组、低级别胶质瘤(Ⅰ~Ⅱ级)、Ⅲ级胶质瘤和Ⅳ级胶质瘤中的表达逐渐增加,差异均有统计学意义(P〈0.05);Western blotting和免疫荧光染色结果显示NOX在对照组标本中呈低表达.在胶质瘤中呈高表达,且胶质瘤恶性程度越高,表达水平越高。结论NOX的表达及ROS含量与胶质瘤的恶性程度有关,可能对胶质瘤的发生和恶性增殖起重要作用。Objective To detect the expression ofNAD(P)H oxidase (Nox) and the content of RIDS in different malignant gliomas, and explore the influence of Nox and intracellular ROS levels in the survival, proliferation, and malignant phenotype of gliomas. Methods Thirty human glioma specimens (10 with grade I and II, 10 with grade III and 10 with grade IV), performed resection in our hospital from August 2007 to August 2010, were collected in our study; another 10 normal brain tissues were collected as controls. Real time PCR was used to detect the mRNA expressions ofNox(1-5); ROS level was detected by flow cytometry and Nox4 protein level was detected by immunofluorescence and Western blotting. Results The Noxl-5 mRNA and protein levels and ROS content were significantly different between each 2 groups (P〈0.05); Nox4 mRNA expression and ROS level significantly increased following the increment of malignancy degrees (controls〈low-grade gliomas〈grade III gliomas〈 grade IV gliomas,/~〈0.05); the Nox protein expression was low in controls while that in gliomas was high, and the higher malignancy of the tumors, the higher expression levels of the tumors. Conclusion Nox expression and ROS content have significantly positive relations with the malignancy of the tumors, which indicates that Nox expression and ROS content might paly important roles in the occurrence of glioma and malignant proliferation.
