摘 要:目的通过阶段性观察大蒜素处理后Wistar大鼠诱癌模型中异常腺窝病灶(ACF)的发展情况,比较和评价大蒜素对肠道病变的预防作用。方法 80只Wistar大鼠给予二甲肼(DMH)皮下注射(20 mg/kg,每周1次,连续18周),分为大蒜素处理组和非处理组,每组分两批,分别于第8、16周处死,将美蓝染色后的大肠组织在实体镜下观察。结果 ACF均分布于中段及远端大肠(大部分位距盲肠50%~80%处),共发现肿瘤2个,均为高分化腺癌。大蒜素处理组ACF数目较非处理组明显减少,差异有显著性统计学意义。结论大蒜素可以明显减少Wistar鼠诱癌模型中异常腺窝病灶的数量,从而进一步证实其对结直肠癌的预防作用。Objective To evaluate the development of aberrant crypt loci (ACF) to tumor in the DMH-treated rats treated by the allicin. Methods 80 Wistar rats were given dimethyl hydrazine (DMH) subcutaneous injection (20 rag/ kg weekly for 18 weeks) , they were divided into allicin treated group and non-treated group, and these rats were sacri- ficed in 8, 16 weeks respectively. The methylene blue staining of colorectal tissue was observed in the platoscope. Re- sults ACF located in the middle and distal colon (most of the ACF located far from the cecum 50% to 80% ) , two tumors well-differentiated adenocarcinoma were identificated by histology. The significant difference of ACF development was found between allicin treated group and non-treated group. Conclusion Allicin can significantly reduce the devel- opment of aberrant crypt foci in Wistar rats, which is further certificated to play the role in the prevention of colorectal cancer.
