摘 要:为定量测定酶法合成龙胆低聚糖得率,采用葡萄糖氧化酶-过氧化物酶法(GOD-POD),通过测定转化后残留底物葡萄糖含量,间接测定龙胆低聚糖得率。对该法测定酶法合成龙胆低聚糖体系中残留底物葡萄糖含量的干扰因素、方法的稳定性、重复性、精密度进行研究。结果:该法在2h内测定体系葡萄糖含量结果稳定(RSD=0.81%),重复性好(RSD=0.97%),精密度高(RSD=1.55%),干扰小(加样回收率=101.681%)。结论:GOD-POD法用于测定酶法合成龙胆低聚糖体系中残留底物葡萄糖含量结果稳定可靠,体系中产物龙胆低聚糖及其他物质对GOD-POD法测定葡萄糖含量无显著影响。用该法间接测定酶法合成龙胆低聚糖得率可行。In order to determine yield of Gentiooligsaccharide quantitatively when it was prepared by enzyme, the Oxidase-Peroxidase (GOD-POD) method was used to determine content of residual substrate glucose, and to measure yield of Gentiooligsaccharide indirectly. The interference factors of GOD-POD method for determining content of residual substrate glucose in system of Gentiooligsaccharide prepared by enzyme, the stability, repeatability, and precision of the method were all studied. It was showed the results of determining content of residual substrate glucose by GOD-POD method were stable in two hours (RSD=0.81%), with good repeatability (RSD=0.97%), high precision(RSD=1.55%),and slight inference (the average recovery=101.681%), indicating that the results of determination yield of Gentiooligsaccharide were stable and trustworthy. Gentiooligsaccharide and other kinds of materials did not affect the determination notably. It was available to use GOD-POD method to determine indirectly yield of Gentiooligsaccharide prepared by enzyme.
