摘 要:目的:以新疆和田维吾尔族长寿老人和一般人肠道中筛选出来的27株菌株为试材,采用耐胆汁盐,耐酸,降胆固醇等实验方法对该菌株进行综合性分析,并对其降胆固醇机理进行初步探讨。方法:采用硫酸铁铵法测定该菌株对培养基中胆固醇的脱除率,同时用吸光度法对候选菌株耐酸,耐胆汁盐以及蛋白酶的耐受性进行检测,筛选综合性能高的菌株。结果:27株实验菌都能从培养基中除去胆固醇,脱除率在22.32%~78.57%之间,平均脱除率50.63%,13株脱除率高于平均值(50.63%)。其中10株菌株脱除率高于52.58%,被选为后续研究的菌株;又通过胆汁盐耐受性,低pH耐受性和蛋白酶耐受性实验,从10株菌中筛选出综合性能比较高的R02-23,R03-13,S04等3株菌。结论:从长寿老人肠道中分离筛选的R02-23,R03-13,S04菌株均具有降胆固醇,耐胆汁盐,耐酸,耐蛋白酶能力。且胆固醇的降解机理研究表明它们都有吸收胆固醇和同化胆固醇的能力,但对病原菌的抑菌作用机理有待进一步研究。Objective ; 27 strains Entrocuccus isolated from gut of long-lived persons and general people from Hotan,Xinjiang,were analyzed respectively by conducting the experiment of removal cholesterol from media, bile-tolerance and acid tolerance,and preliminarily researched on cholesterol lowering mechanism in order to find out the relationship between probiotics and longevity.Methods;Cholesterol removal rate was measured on culture medium of strains by using ammonium ferric sulfate,and screened strains which has high comprehensive performance and studied the strains of cholesterol lowering mechanism by testing degree the of bile-tolerance, acid-tolerance and protease-tolerance through the absorbance of candidate strains. Results,,All of the 27 strains of experimented bacteria can remove cholesterol from the culture medium,the removal rate wasbetween 22.32%-78.57% and the average removal rate was 50.63%, in addition, removal rate of 13 strains was higher than average rate(50.63%). The 10 strains of which removal rate was higher than 52.58% were selected to extend study. The strains of RO2-23,R03-13 and SO4 were selected as the strains of high comprehensive performance by using tolerance tests of bile salt,low pH and protease. The research about cholesterol degradation mechanism of these bacteria showed that they had the ability of cholesterol absorption and cholesterol assimilation. However,the pathogen antibacterial mechanism of the strain was needed further study. Conclusion.Isolated strains RO2-23,R03-13 and SO4 were with the characteristics of bile-tolerance,acid- tolerance and protease-tolerance from gut of long-lived beings. It could be clearly showed from the cholesterol lowering mechanism that they could absorb and assimilate cholesterol, but its inhibiting mechanism in pathogenic bacteria was needed to further studv.
