摘  要:在富铬Z氏培养液的条件下,研究了钝顶螺旋藻对Cr(Ⅲ)的吸收及其有机化程度,采用不同饱和度硫酸铵沉淀的方法分离提取不同蛋白组份,对其含Cr(Ⅲ)量进行了分析,并探讨了蛋白组份对α-葡萄糖苷酶活性的抑制作用。结果表明,螺旋藻培养液中CrCl3.6H2O添加浓度为300 mg·L-1,螺旋藻总铬富集量可达到3.30 mg·g-1,有机化程度为70.2%;不同饱和度(25%、50%和75%)硫酸铵沉淀出的蛋白组份中铬占总铬的情况分别为2.1%、16.8%和7.0%,不同蛋白质结合铬的能力各异;3种蛋白组份对α-葡萄糖苷酶均有抑制作用,其中50%饱和度硫酸铵沉淀的蛋白组份抑制活性最高,IC50为69.87 mg·mL-1。In this paper, we studied the absorption degree of Cr(Ⅲ) in Spirulina platensis which cultivated in chromium (Ⅲ)-riched Zarrouk medium, by extracting proteins with ammonium sulfate precipitation, analYzing the content of Cr(Ⅲ) in protein components. And then we investigated the inhibition effect of chromium (Ⅲ)-riched Spirulina platensis proteins on alpha-glucosidase. The results showed that Spirulina platensis growed in,culture medium added with 300 mg·L^1CrC13·6H2O, the total enriched Cr(Ⅲ) and it,; organic degree of Cr(Ⅲ) could reach to 3,30 mg·g^-1 and 70.2%, correspondingly. When different saturated solution (25%, 50% and 75%) of ammonium sulfate were used to precipitate proteins, the proportion of Cr(Ⅲ) in protein components to total Cr(Ⅲ) were 2.1%, 16.8% and 7.0%, respectively. The abilities of different proteins combined with Cr (Ⅲ) were different. The three protein components had different inhibition activities on alpha-glucosidase. Among them, protein com- ponents precipitated by 50% saturation solution of ammonium sulfate revealed the best inhibition effect on al- pha-glucosidase, with IC50 of 69.87 mg·mL^-1.
