摘  要:目的探讨中链脂肪酸(MCFA)对KKAy糖尿病小鼠脂代谢的影响及相关机制研究。方法 36只4周龄KKAy雄性糖尿病小鼠,随机分为3组,每组12只,分别给予高脂饲料添加辛酸(1.5%,C8)、高脂饲料添加癸酸(1.5%,C10)、高脂饲料添加油酸(1.5%,OA),喂养12周。观察各组小鼠体重及体脂肪质量,检测血清甘油三酯(TG),胆固醇(TC),游离脂肪酸(FFA)、脂蛋白脂酶(LPL)、脂肪酸合成酶(FAS)、乙酰CoA羧化酶(ACC)。结果喂养12周后,辛酸组体质量(42.84±1.57)g和癸酸组体质量(42.19±1.64)g均显著低于油酸组(45.60±0.77)g,(P<0.05);辛酸组、癸酸组小鼠肠系膜周围、附睾周围和肾周围脂肪垫重显著低于油酸组(P<0.01或P<0.05);辛酸组和癸酸组TG,TC,LDL-C、FFA水平显著低于油酸组(P<0.05),辛酸组和癸酸组LPL、FAS、ACC活性较油酸组降低(P<0.05)。结论辛酸、癸酸可显著减少糖尿病小鼠体重和体脂肪增加,其途径可能与中链脂肪酸可降低脂肪酸合成酶活性并提高脂肪酸氧化分解酶活性等有关。Objective To study the effect of medium chain fatty acid on lipid metabolism of mice with diabetes mellitus(DM) and its mechanism. Methods Thirty-six male kkay DM mice were randomly divided into 3 groups(12 in each group). After they were fed with high fat diet containing octoic acid(1.5%, C8), capric acid(1.5%, Cl0)and oleinic acid(1.5%, OA), respectively, for 12 weeks, their body weight and body fat, and triglyeride(TG), cholesterol(TC), free fatty acid(FFA), lipoprotein lipase(LPL), fatty acid synthetase(FAS)and acetyl-CoA-carboxylase(ACC) levels were measured. Results Twelve weeks after the mice were fed with octoic acid, capric acid and oleinic acid, their body weight and body fat around mesenterium, epididymis and kidney, and LPL, FAS, ACC, TG, TC, LDL-C and FFA levels were significantly lower in octoic acid group and capric acid group than in oleinic acid group(42.84± 1.57)g and (42.19 ± 1.64)g vs (45.60 ± 0.77)g, (P〈0.05). Conclusion Octoic acid and capric acid can significantly reduce the body weight and body fat of DM mice by decreasing the activity of synthetase and increasing the activity of oxidase in medium chain fatty acid.
