晚期癌症的治疗策略:从靶向治疗到基因治疗

摘　　要：本文作者基于对白血病等恶性肿瘤发病原理和病因的系统认识和国外最新成果,在对早中期癌症基因治疗成功的基础上,提出晚期癌症的治疗策略,即从癌蛋白和癌基因两个层面上采用两种方案即:①靶向三个癌基因(V-erbB,TS和C-Met),它们分别是癌症致病基因,癌变基因和晚期癌基因)产物的小分子抑制剂和5-氟脲嘧啶核苷(5-Fu);②反基因V-erbB+CpG双寡核苷酸组合方案,相结合。首先采用:①方案消灭大量癌细胞,再用②方案,针对癌干细胞和残余癌细胞进行综合治疗。继续使用第二方案,直到完全缓解5年以上。上述治疗可称为基因产物治疗和基因治疗。希望医者在实践中加以优化和完善。Based on our understanding of mechanism of leukomo-carcinogenesis and on recent progressions of cancer invasive,metastastic and signal transduction pathway investigations in literatures and also on successes in gene therapy for rat MDS and early esopheagical cancer,author present his therapeutic strategy for advanced cancers that include 2 scheme : the first,patients will be administrated 3 oncogene(V-erbB,C-Met and TS) product inhibitors,such as TKIs and 5-Fu for treatment of the big count cancer cells;and then patients will be used second scheme that include two oligodeoxynucleotides(antigene V-erbB and CpG ODN) for treatment of cancer stem cells and residual cancer cells in order to prevention of cancer relapse and metastasis.Third,patient will be continue to be administrated two ODNs upto more than 5 years.This therapy could be named as Gene Product Therapy and Gene Therapy as well.
