摘 要:建立高效液相色谱法测定枸杞叶粗蛋白中活性多肽IN-5含量的方法。色谱条件为:色谱柱Kinetex C18(250mm×4.6mm,2.6μm);流动相为乙腈-0.2%磷酸水(5∶95,V/V);检测波长210nm;流速为0.7mL/min;柱温25℃。结果表明,IN-5浓度在16.0—240.0μg/mL内与峰面积的线性关系良好(r=0.9999)。平均回收率99.2%,RSD=0.49%(n=9)。方法快速、准确、重复性好,可用于IN-5含量的检测。The method for determination of the content of active hypoglycemic peptide IN-5 in the crude protein extract of Lycium chinense leaves was established by HPLC.The chromatographic conditions were as follows:Kinetex C18 column(250mm×4.6mm,2.6μm),mobile phase of acetonitrile-0.2% phosphoric acid(5∶95,V/V),detection wavelength of 210nm,flow rate of 0.7mL/min and column temperature of 25℃.The results showed that the concentration of IN-5 had a good linear relationship with peak area(r=0.9999) between 16.0μg/mL and 240.0μg/mL.The average recovery was 99.2% with RSD of 0.49%(n=9).The method is fast,accurate with good repeatability,and can be applied to detection of the content of IN-5.
