摘 要:食用植物油经深度氧化劣败会产生高浓度的氧化三酰甘油聚合物(TGP),TGP是氧化三酰甘油(oxidized triacylglycerol,ox-TG)的二聚体(oxidized triacylglycerol dimers,TGD)和寡聚体(oxidized triacylglycerol oligomers,TGO)的总称。研究旨在初步评价TGD和TGO的生物学毒性。以不同浓度的TGP作用于体外培养的巨噬细胞,分别检测细胞的增殖能力、中性红吞噬能力和LDH释放量,同时采用激光共聚焦显微镜观察细胞凋亡、流式细胞术检测细胞内ROS水平。结果显示,TGP显著抑制巨噬细胞的增殖能力和中性红吞噬能力,显著提高细胞内ROS水平和LDH释放量,导致巨噬细胞的凋亡。因此,初步证实TGP具有明显的生物学毒性,对机体的免疫功能有损伤作用。Edible vegetable oil usually produces high concentrations ofoxidized triglyceride polymers (TGP) after deeply oxidative deterioration. The aim of the present study is to evaluate the toxicity of TGP. Cultured macrophages were treated with different concentrations of TGP and then the cell proliferation ability, the phagocytic activity and LDH release were detected. Besides, apoptosis were observed by laser scanning confocal microscopy, and ROS were determined by flow cytometry. The results showed that proliferation ability and phagocytic activity of macrophages were significantly inhibited by TGP. TGP also significantly increased the ROS level and LDH release of cells, leading apoptosis in macrophages. Therefore, it is suggested that TGP has obvious toxicity and may damage the immune function.
