摘 要:目的:观察表没食子儿茶素没食子酸酯(EGCG)对肥胖幼鼠模型动脉病变和水通道蛋白7mRNA表达的干预作用。方法:通过过度喂养建立食饵性肥胖幼鼠模型,将出生第3天的雄性SD大鼠72只随机分为对照组、模型组、EGCG低剂量组和EGCG高剂量组,每组18只,HE染色观察各组腹主动脉早期动脉病变,同时运用real-time PCR测定脂肪和腹主动脉组织AQP-7mRNA表达。结果:EGCG低剂量可降低腹主动脉平滑肌中粥样斑块形成,EGCG高剂量可有效改善腹主动脉早期动脉粥样硬化形成,下调脂肪组织和腹主动脉AQP-7mRNA表达水平(P<0.01)。结论:EGCG可能通过下调肥胖大鼠脂肪组织和腹主动脉AQP-7mRNA表达水平而起到减肥和改善早期动脉粥样硬化的作用。Objective: To investigate the effect of epigallocatechin-3 gallate(EGCG) on early arterial disease and the expression of aquaporin 7(AQP-7)mRNA in obese young rats.Methods: Obese model was induced by over-feeding in young SD rats.A total of 72 3-day young rats were randomly divided into control group,model group,low-dose EGCG group,and high-dose EGCG group(18 in each).Early abdominal aortic artery lesions were observed by HE staining.AQP-7 mRNA expression in fat and abdominal aorta tissues was determined by Real-Time PCR.Results: Lose-dose EGCG reduced the formation of atherosclerotic plaques in the abdominal aorta smooth muscles;high-dose EGCG effectively improved attenuated early atherosclerosis of the abdominal aorta and at the same time downregulated the AQP-7 mRNA expression in fat tissue and abdominal aorta as compared to the model group(P〈0.01).Conclusions: EGCG may control the weight and attenuate early atherosclerosis of the abdominal aorta by decreasing the level of AQP-7 mRNA in adipose tissue and abdominal aorta in young obese rats.
