摘 要:目的探索通过盐效应分离长孢葡萄穗霉FG216(Stachybotrys longispora 216)发酵液中纤溶活性化合物FGFC1(fungi fibrinolytic compound 1)的方法。方法长孢葡萄穗霉FG216经过种子液培养和发酵培养获得的发酵液经浓缩后添加氯化钠、氯化铵和氯化钙至不同饱和度,用乙酸乙酯萃取,通过HPLC检测FGFC1的分离效果,并分析FGFC1的质谱和光谱特性。结果随着氯化钠、氯化铵和氯化钙的饱和度从20%升高到90%,氯化钠和氯化钙的盐效应分离作用效果呈现逐渐增强的趋势,60%饱和度的氯化钠能达到18.8 mg/mL的FGFC1提取率,是甲醇分离方法的118%,采用盐效应分离和用甲醇分离获得的FGFC1的质谱和光谱特性一致。结论用氯化钠盐效应分离长孢葡萄穗霉FG216发酵液中FGFC1是一种有效的分离方法,该盐效分离结果符合盐效分离的基本理论。Purpose To explore the isolation method of fibrinolytic compound FGFC1 (fungi fibrino- lyric compound 1 ) by salt effect from the broth of Stachybotrys longispora FG216. Methods The broth was concentrated after the salts including sodium chloride, ammonium chloride and calcium chloride were add- ed ,respectively, following that the concentrated liquor was extracted with ethyl acetate, The separation effect of FGFC1 was determined by HPLC,mass spectrometry and spectral characteristics. Results Exper- iment with the different saturations of sodium chloride, ammonium chloride and calcium chloride increased from 20% to 90%. The isolation effect of sodium chloride and calcium chloride showed a gradual increas- ing trend. When the saturation of sodium chloride was 60% ,the extraction rate of FGFC1 can reach 18.8 mg/mL,which accounted for 118% of methanol extract. The mass and spectral characteristics of FGFC1 are consistent with those extracted by salt effect and extracted by methanol. Conclusion Isolating fibrino- lytic compound FGFC1 by salt effect is an effective method of separation and the salt separation efficiency accords with the basic theory of salts effective separatian.
