摘  要:以单宁微球为载体制备固定α-淀粉酶.探讨了温度、pH值、淀粉初始质量浓度、淀粉种类等对单宁微球固定化α-淀粉酶催化淀粉水解性能的影响.结果表明,固定α-淀粉酶在25,60,90℃催化水解淀粉17h后,淀粉的水解百分率均达到95%以上,说明温度对固定α-淀粉酶的催化性能影响不大;pH对固定α-淀粉酶的催化性能影响大,最佳pH值为8;固定α-淀粉酶对可溶性淀粉、番薯淀粉、芭蕉芋淀粉的催化水解率均大于96%;固定α-淀粉酶重复5次后仍具有较好的催化效果.α-amylase was immobilized on tannin microspheres. The effects of tempera-ture, pH and initial concentration of starch on the catalytic performance of immobilized en-zyme were investigated. The results showed that each of the hydrolysis degree of starch hy- drolyzed by immobilized a-amylase under 25,60 and 90 ℃ for 17 h was more than 95%. The effect of temperature on catalytic performance of the immobilized α-amylase was limited. The optimum of pH was 8. The hydrolysis degree of soluble starch, sweet potato starch and can-na edulis starch, which was hydrolyzed by immobilized α-amylase under 60 ℃ for 17 h, was more than 96%. Moreover, the immobilization enzyme still had a good catalytic performance on hydrolysis of starch after having been reused repeatedly 5 times.
