摘 要:以短乳杆菌为出发菌株,采用大气压冷等离子体介质阻挡放电法和离子束对乳酸菌进行诱变。以菊芋作为筛选培养基,摇瓶筛选得到高产甘露醇的菌株。之后对筛选出的高产甘露醇菌株进行遗传稳定性实验,证实该菌株遗传稳定。在5L发酵罐中进行发酵,该菌株可有效地将果糖转化为甘露醇,产率达到62%。这株诱变菌可作为高产甘露醇的优良菌株。Mannitol is the main product when fructose is used as the substrate for heterofermentative lactic acid bacteria under anaerobic conditions. In this study, the mutation of a Lactobacillus brevis strain was induced by atmospheric pressure dielectric barrier discharge plasma and ion beam to im- prove its mannitol productivity. A positive mutant was selected for its superior mannitol productivity when saccharified Jerusalem artichoke juice was used in selective medium. And it was also confirmed that the mutant strain had high genetic stability. When fermented in 5L fermentor, the mutant could effectively convert fi'uctose into mannitol and the yield of marmitol reached 62%. It was indicated that the mutant strain could be a candidate for industrial mannitol production.
