摘  要:目的探讨嗅成鞘细胞(OECs)与神经干细胞(NSCs)共移植对阿尔茨海默病(AD)大鼠海马生长相关蛋白(GAP)-43表达的影响。方法雄性SD大鼠双侧穹隆-海马伞切断,建立AD大鼠模型。实验动物分为4组:NSCs移植组、NSCs与OECs共移植组、AD模型组、正常对照组,每组30只。从SD胎鼠(孕16~18d)取材,在体外分别培养和共培养OECs与NSCs,并将共培养的OECs与NSCs移植于AD大鼠大脑侧脑室;观察各组大鼠行为学、宿主脑海马GAP-43和NOS阳性神经元表达的变化。结果①NSCs与OECs共移植组潜伏期明显缩短、通过平台次数明显增加,与NSCs移植组、AD模型组差异有统计学意义(P<0.05)。②反转录聚合酶链反应(RT-PCR)与Western Blot分析,NSCs与OECs共移植组宿主脑海马GAP-43mRNA和GAP-43蛋白的表达量高于NSCs移植组、AD模型组(P<0.05)。③NSCs与OECs共移植组NOS阳性细胞(22.2±1.7)较NSCs移植组(13.1±1.9)、AD模型组(7.1±0.9)明显增多(P<0.05)。结论嗅成鞘细胞与神经干细胞共移植能促进宿主脑海马GAP-43的表达,其可能在AD大鼠中枢神经系统损伤时具有促进神经再生和修复的作用。Objective To investigate the effect of co-transplantation of Olfactory Ensheathing Cells(OECs) and Neural Stem Cells(NSCs) on growth associated protein-43(GAP-43) expression in the hippocampus for rats' Alzheimer Disease(AD). Methods To establish AD SD rat model by transecting bilateral fimbria fornix of adult SD male rats. The experimental animals were divided into 4 groups, namely, NSC transplantation group, cotransplantation of NSCs and OECs group, AD model group and control group, with 30 rats in each group. SD fetal rats (pregnant for 16--18 d) were selected; OECs and NSCs were cultured in vitro, respectively, and OECs were co-cultured with NSCs. Then the co-cultured OECs and NSCs were transplanted into cerebral ventricle of AD rats, in order to observe the changes of the rats' behaviors, as well as the expression of GAP-43 and NOS positive neurons in the hippocampi of host brain. Results ①The latency of the co-transplantation of NSCs and OECs group was considerably reduced, with significant increase of the number of passing the platform. The co-transplantation group also had significant difference with NSC transplantation group and AD model group, respectively (P〈 0.05) ; ②RT-PCR and Western Blot analysis showed that co-transplantation of NSCs and OECs group rats had a significantly higher GAP-43 expression in the hippocampus compared with NSC transplantation group and AD model group (P〈0.05) ; ③The number of NOS positive cells in co-transplantation of NSCs and OECs group (22.2±1.7) was significantly higher than that in NSC transplantation group (13.1± 1.9) and AD model group (7.1±0.9) (P〈0.05). Conclusion Co-transplantation of OECs and NSCs may obviously promote GAP-43 expression in the hippoeampus for rats'AD.
