摘 要:目的研究基因工程抗菌肽对肿瘤细胞的选择性抗肿瘤作用及对大鼠大肠肿瘤的影响。方法采用琼脂扩散法检测基因工程抗菌肽生物活性及致死浓度(LC),MTT法测定对人胃黏膜上皮细胞株GES-1、人结肠癌细胞株LS-174T的细胞毒作用及半数抑制浓度(IC50)。以二甲肼(DMH)诱发的Wistar大鼠大肠肿瘤为模型,观察基因工程抗菌肽对大鼠大肠肿瘤的防治作用。结果基因工程抗菌肽及对照抗菌肽的致死浓度(LC)分别为1.87μmol/L和0.44μmol/L。基因工程抗菌肽、对照抗菌肽及5-FU对胃黏膜上皮细胞株GES-1的IC50分别为大于64.0、60.0和84.0μmol/L,对人结肠癌株LS-174T的IC50分别为10.3、20.0和63.0μmol/L。基因工程抗菌肽灌胃明显降低大鼠结肠肿瘤发生率(58%,P<0.05),大肠肿瘤平均个数及肿瘤质量指数(TMI)差异无统计学意义。结论基因工程抗菌肽在一定浓度范围内对肿瘤细胞具有选择性杀伤作用,动物实验结果提示可防治DMH诱导的大鼠大肠肿瘤发生。【Objective】 The aim of the present work was to gain insight into a selective anticancer effect of gene engineering antibacterial peptide on cancer cells and rat colorectal cancer model.【Methods】 The inhibition zone assay was used to determine the biological activity and lethal concentration(LC) of gene engineering antibacterial peptide.The cytotoxicity assay and half-inhibitory concentration(IC50) was undertaken on human gastric epithelial cell line GES-1 and human colon adenocarcinoma cell line LS-174T by MTT(microculture tetrazolium) colorimatric methods.The gene engineering antibacterial peptide was investigated the preventive and therapeutic effects in Wistar rat colorectal cancer model induced by 1,2-dimethylhydrazine.【Results】 The lethal concentration for gene engineering antibacterial peptide and control were 1.87 μmol/L and 0.44 μmol/L.The IC50 for the gene engineering antibacterial peptide,control and 5-Fluorouracil(5-FU) on human gastric epithelial cell line(GES-1) were over 64,over 60 and 84 μmol/L,and for human colon adenocarcinoma cell lines(LS-174T) were 10.3,20.0 and 63.0 μmol/L.There was a significantly lower colorectal tumor incidence in rat cancer model gavaged gene engineering antibacterial peptide(58%,P 0.05).The tumor burden(tumors/tumor-bearing animal) and tumor mass index were not different in the groups.【Conclusions】 The gene engineering antibacterial peptide has tumor-specific cell killing effects in colorectal carcinoma cells at certain concentration.The animals test results indicate that gene engineering antibacterial peptide impedes the neoplastic process in rat induced by 1,2-dimethylhydrazine.
