摘 要:目的探讨骨髓移植前供鼠经口服受鼠脾淋巴细胞诱导免疫耐受对于预防移植物抗宿主病(GVHD)的作用。方法用主要组织相容性抗原(MHC)完全不合的纯种近交系小鼠[供鼠:雄性C57BL/6J(H-2b)鼠;受鼠:雌性BALB/c(H-2d)鼠]建立异基因骨髓移植(allo-BMT)GVHD动物模型,随机分为4组(每组10只):A组为口服耐受组,移植前供鼠用受鼠脾淋巴细胞喂养;B、C组为受鼠在移植后分别使用雷帕鸣、环孢菌素A+甲氨蝶呤;D组不使用免疫抑制剂作为模型对照。ELISA法检测各组小鼠骨髓移植后细胞因子水平的差异。结果 A组小鼠IL-2、IL-6、IL-8、TNF-α水平较B、C、D组明显减低,且增长较其亦明显缓慢(P<0.05或P<0.01);而IL-10水平较B、C、D组增高(P<0.05)。结论供鼠口服耐受可明显抑制受鼠GVHD,其效果优于传统免疫抑制药物。Objective To evaluate the inhibition effect of feeding the donors with recipient splenocytes before allogeneic marrow transplantation (allo-BMT) on graft-versus-host-disease (GVHD) in mice model. Methods The allo-BMT/GVHD models were established in 40 mice, which were equally randomized into 4 groups of A (the dornors fed with recipient splernocytes before allo- BMT), B(given rapamycin after allo-BMT), C(given ciclosporin A plus methotrexate after allo-BMT) and D (the recipient mice given nothing after allo-BMT as model control). The GVHD-related cytokines were detected with ELISA. Results The levels of IL-2, IL-6, IL-8 and TNF-α were significantly lower, but IL-10 was higher, in group A than those in groups of B, C and D(P〈0. 05). Conclusion In allo-BMT in mice, oral tolerization of the donors can significantly inhibit GVHD of the recipient mice,which is better than conventional immunosuppressive drugs.
