摘  要:目的:通过分析肝细胞癌(HCC)基因表达谱改变来探讨HCC发病机制。方法:从GEO数据库中下载HCC基因表达数据GSE14215和GSE29217,采用Genespring 11.0软件分析差异表达基因,运用统合分析(meta-analysis)方法确定共同差异表达基因,利用DAVID、KEGG、String和FABLE等生物信息学软件确定相关的生物学过程、分子功能、相互作用通路等。结果:HCC组织与正常肝组织相比,共同下调、上调的差异表达基因分别是281和227个,其中作为转录因子36个;与文献报道的HCC相关基因一致的有181个。差异表达基因主要与细胞周期调控、代谢调节、药物代谢酶系、DNA的损伤复制、p53和过氧化物酶体增殖物激活受体信号通路(PPAR)等信号通路有关。结论:HCC基因表达谱数据分析表明,细胞周期紊乱主要发生在S、M期;转录因子FOXM1/HNF3高表达与HCC的细胞周期紊乱密切相关。OBJECTIVE: To investigate the mechanism of hepatocellular carcinoma (HCC) by analyzing the alteration of gene expression profile. METHODS: The common differential expressed genes in HCC were detected with the software of Genespring 11.0 and recta-analysis from the data series of GSE14215 and GSE29217 downloaded from Gene Expression Omnibus (GEO). The biological process, molecular function, signal pathway, protein-protein interaction of the differential genes were created by the bioinformatics tools of DAVID, KEGG, String, FABLE, etc. RESULTS.. There are coincidental 281 up- and 227 down- regulated genes in all three microarray platforms in HCC tissue compared to normal liver tissue or adjacent tumor tissue. Thirty-six genes of which are transcription factors and 181 genes associated with HCC have been reported by the researchers. The differential expressed genes are mainly involved in the regulation of cell cycling and metabolism, the system of drug metabolism, the process of DNA repair and replication, the signal path- way of p53 and peroxisome proliferator-activated receptors signaling pathway (PPAR). CONCLUSION: The results sug- gest that the abnormality of cell cycle could mainly take place in the phase of S and M, and the high expression of FOXM1/HNF3 transfactor could be associated with the cell cycle confusion of HCC.
