摘 要:目的本实验利用长期双侧颈总动脉结扎法制备血管性痴呆(Vascular dementi,VD)大鼠模型,并观察大鼠海马齿状回(Dentate gyrus,DG)区细胞外液中几种氨基酸含量的变化,探讨血管性痴呆记忆认知障碍和海马DG区氨基酸类物质含量的关系。方法将成年Wistar大鼠(250~300 g)随机分成对照组(n=6)、假手术组(n=6)和模型组(VD组)(n=6)。对照组不做手术,假手术组只剥离双侧颈总动脉但是不结扎,模型组剥离并永久性结扎双侧颈总动脉。术后30 d开始采用Morris水迷宫进行学习记忆的行为学观察;训练结束后利用脑内微量透析法和高效液相色谱法测定海马DG区细胞外液中的7种氨基酸类物质(包括天门冬氨酸、谷氨酰胺、谷氨酸、甘氨酸、牛磺酸、丙氨酸和γ-氨基丁酸)的含量。结果 (1)VD组与假手术组、对照组比较,水迷宫实验的逃避潜伏期明显延长(P<0.05),假手术组和对照组之间无明显差异(P>0.05)。(2)VD组海马DG区细胞外液中的谷氨酰胺、牛磺酸、丙氨酸和γ-氨基丁酸的含量均明显高于对照组和假手术组,(P均<0.05)。其余3种氨基酸无明显变化(P均>0.05);假手术组的海马DG区7种氨基酸含量与对照组比较无明显差异(P>均0.05)。结论 (1)采用双侧颈总动脉结扎术能有效制备VD模型;(2)VD可能与海马DG区谷氨酰胺、牛磺酸、丙氨酸和γ-氨基丁酸含量增加有关。Objective To observe the extracellular concentration of several amino acids in hippocampal DG region in vascular dementia rat and to discussed the relationships between memory disorder and the extra- cellular concentration of several amino acids. Methods Adult wistar rats (250-300 g) were randomly divided into control group (n = 6), sham operation group (n = 6) and VD group (n = 6). Operation wasn't received in control group, bilateral carotid arteries were only dissected in .sham operation group, and bilateral carotid ar- teries were occluded in VD group. 30 days after surgery, learning and memory behavior were observed using Morris water maze. After training, the extracellular concentrations of several amino acids(including aspartic acid, glutamine, glutamic acid, glycine, taurine, alanine and γ-aminobutyric acid) in hippocampal DG region were tested by using brain microdialysis and high performance liquid chromatography techniques. Results Compared with the sham group and the control group, water maze escape latency was prolongedin VD group (P〈0. 05) and the extracellular concentrations of glutamine, taurine, alanine and γ-aminobutyric acid significantly increased in VD group (P〈0. 05). There was no significant change in the other three amino acids (P〉0. 05), and there was no significant difference between the control and the sham group in the concentrations of several amino acids (P〉0. 05). Conclusions (1)Bilateral carotid artery ligation can effectively prepare VD model; (2)VD may be associated with increasing of glutamine, taurine, alanine and γ-aminobutyric acid in hippocampal DG region.
