摘  要:目的观察浒苔多糖功能饮料(EB)对2型糖尿病大鼠脂代谢影响及其作用机制。方法高糖高脂饲料喂养,加小剂量注射STZ(30 mg/kg)造模,EB灌胃干预28 d。测定大鼠周空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和胰岛素(INS)水平并计算胰岛素敏感指数(ISI),检测大鼠肝脏组织中葡萄糖激酶(GCK)、胰岛素受体(InsR)和脂肪组织中葡萄糖转运体(GLUT)-4、脂联素(APN)mRNA的表达水平。结果与模型组比较,EB各剂量组FBG和INS较低,ISI较高,EB高、中剂量组TC、TG、LDL-C均明显较低,EB高、低剂量组MDA较低、SOD和GSH-Px均较高,EB各剂量组肝脏InsR mRNA的表达较低,脂肪GLUT-4 mRNA表达较高,EB高、中剂量组肝脏GCK mRNA和脂肪APN mRNA表达量均较高(P<0.05或P<0.01)。结论 EB可通过上调脂肪组织中APN的表达,从而增强胰岛素敏感性,改善IR;也可通过上调脂肪组织GLUT-4和肝脏组织葡萄糖代谢关键酶GCK基因的表达,促进葡萄糖的转运和代谢,改善糖脂代谢异常。Objective To observe the effects of polysaccharides in enteromorpha functional beverage (EB) on adjusting the glyco- lipid metabolism of type 2 diabetic (T2DM) rats and discuss its possible mechanisms. Methods Rats model of T2DM were established by a high sugar and fat diet with intraperitoneal injection of STZ (30 mg/kg), and then given various doses of EB for 28 days. Fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein choles-terol ( LDL-C), malondialdehyde ( MDA), superoxide dismutase ( SOD), glutathione peroxidase ( GSH-Px), insulin (INS) were measured weekly and insulin sensitive index (ISI) was calculated. Expressions of glucokinase (GCK) mRNA, insulin receptor (InsR) mRNA in liver tissue and GLUT-4 mRNA, adiponectin (APN) mRNA in fat tissue were detected by RT-PCR. Results Compared with model group, FBG and INS in all EB intervention groups were lower, but ISI was higher. TC, TG and LDL-C in high and medium dose EB groups were reduced significantly. MDA was lower, SOD and GSI-I-Px were higher in high and low dose groups. Expression of InsR mRNA in liver was lower and GLUT-4 mRNA in adipose was higher in all EB intervention groups. Expressions of APN mRNA in adipose and GCK mRNA in liver were higher in high and medium dose groups (P 〈 0. 05 or P 〈 0. 01 ). Conclusions EB can up-regulate APN expression in adipose to increase insulin sensitivity, improve the IR, also can up-regulate GLUT-4 and GCK expressions in adipose and liver tissue to promote the transport and metabolism of glucose.
