P16、MCM7、cyclinD1在乳腺癌中的表达及其意义

摘　　要：目的探讨P16、MCM7和cyclinD1在乳腺癌中的表达及意义。方法选取术前未经放化疗的乳腺癌手术标本70例、15例乳腺纤维腺瘤、15例瘤旁正常乳腺组织,应用免疫组化SP法检测P16、MCM7和cy-clinD1基因蛋白的表达,并将70例乳腺癌标本按临床分期和有无淋巴结转移进行分组比较,分析其与乳腺癌临床病理特征的关系。结果乳腺癌中P16阳性表达率47.1%明显低于乳腺纤维腺瘤(86.7%)和瘤旁正常乳腺组织(93.3%)(P<0.01);MCM7阳性表达率85.7%明显高于乳腺纤维腺瘤(13.3%)和瘤旁正常乳腺组织(6.7%)(P<0.01);CyclinD1阳性表达率64.3%明显高于乳腺纤维腺瘤(33.3%)和瘤旁正常乳腺组织(26.7%)(P<0.01)。MCM7和CyclinD1的表达与P16呈负相关(P<0.01)。P16、MCM7的表达均与乳腺癌临床分期、淋巴结转移有关。CyclinD1的表达与临床分期无关,与淋巴结转移有关。结论 P16、MCM7和CyclinD1与乳腺癌的发生、发展有关,三种基因表达联合检测为乳腺癌治疗、预后评估提供依据。【Objective】 To study the expression and significance of P16,MCM7,cyclinD1 in breast cancer.【Methods】 70 cases of breast cancer,15 cases of mammary gland fiber adenoma,15 cases of peritumoral normal breast tissue,were collected from the patients who had not received chemotherapy or radiotherapy before operation.Immunohistochemical method SP was used to detect P16,MCM7,Cyclin D1 gene protein expression.70 cases of breast cancer were grouped according to the clinical staging and lymph node metastasis,and the correlations of the expression levels of P16,MCM7,cyclin D1 in breast cancer with the clinicopathologic features of breast cancer were analyzed.【Result】 Breast cancer P16 positive expression rate of 47.1% was significantly lower than the mammary gland fiber adenoma（86.7%）,peritumoral normal breast tissue（93.3%）,P 0.01;MCM7 positive expression rate of 85.7% was significantly higher than the mammary gland fiber adenoma（13.3%）,peritumoral normal breast tissue（6.7%）,P 0.01;Cyclin D1 positive expression rate of 64.3% was significantly higher than the mammary gland fiber adenoma（33.3%）,peritumoral normal breast tissue（26.7%）,P 0.01.MCM7,cyclinD1 expression and P16 was negative correlation（P 0.01）.The expression of P16,MCM7 were relevant of the clinical stage with breast cancer,the lymph node metastasis.The expression of CyclinD1 was associated with the lymph node metastasis and had nothing to do with breast cancer staging.【Conclusion】 P16,MCM7,cyclinD1 and breast cancer are related to development,united detection P16,MCM7 and cyclinD1 expression provide the basis for treatment,prognosis assessment and follow-up of breast cancer.
