人β防御素2重组蛋白体外抑菌实验及抗肿瘤细胞毒性研究

摘　　要：目的检测重组人β-防御素2蛋白的生物活性,为今后重组人β-防御素2蛋白应用临床提供实验支持。方法将纯化和肠激酶酶切获得的HBD-2,对不同菌株和培养的口腔癌KB细胞进行活力测定,观察基因工程表达纯化获得的HBD-2对不同菌株和口腔癌KB细胞的抑制作用。结果抑菌实验结果表明,HBD-2对不同菌株有不同程度的生长抑制作用,并且抑菌环的大小与剂量有关。HBD-2对人口腔癌KB细胞的生长有明显的抑制作用。结论 HBD-2对大肠杆菌BL21、金黄色葡萄球菌、沙门氏菌、巴氏杆菌均能形成抑菌环,并且抑菌作用与浓度相关。HBD-2对体外培养的口腔癌KB细胞有抑制作用。Objective To detect biological activity of humanβ defensin-2 （HBD〉2） protein for providing experimen tal support for clinical application. Methods The purified and enterokinase digested HBD-2 was used to detect the bio logical activity of various strains and cultural oral carcinoma eukaryotic cell. To observe the inhibiting effect of HBD-2. Results Inhibition zone assays were performed to check the bacteriostasis of purified HBD-2 protein,it had different growth inhibiting effect on different strain. And the size of zone was positive correlate with the protein dose. The growth of oral carcinoma ceil aslo be inhibited by the HBD-2. Conclusion The inhibition zone assays are performed to check the bacteriostasis of purified protein, it has bactericidal activity to Staphylococcus aureus, salmonella,and so on. The size of zone is positive correlate with the protein dose. The growth of oral carcinoma cell aslo can be inhibited by the HBD-2.
