血管紧张素Ⅱ和氧化型低密度脂蛋白对大鼠血管平滑肌细胞Smurf2和Ⅰ型胶原表达的影响

摘  要:目的在体外培养的大鼠血管平滑肌细胞(VSMC)中探讨血管紧张素Ⅱ(AngⅡ)和氧化型低密度脂蛋白(ox-LDL)对Smad泛素调节因子2(Smurf2)及Ⅰ型胶原表达的影响。方法应用RT-PCR检测Smurf2 mRNA的表达,Western blot检测Smurf2蛋白的表达,ELISA检测Ⅰ型胶原的表达。结果与对照组相比,AngⅡ呈浓度依赖性抑制Smurf2 mRNA和蛋白的表达(P<0.05),促进Ⅰ型胶原的表达(P<0.05);ox-LDL呈浓度依赖性促进Smurf2 mRNA和蛋白的表达(P<0.01),抑制Ⅰ型胶原的表达(P<0.05)。以AngⅡ(10-5mol/L)联合不同浓度ox-LDL(20、40及80mg/L)作用大鼠VSMC后,与对照组相比,联合组Smurf2的表达升高(P<0.05),抑制Ⅰ型胶原的表达(P<0.05)。结论 AngⅡ可能通过抑制Smurf2的表达来提高Ⅰ型胶原的表达;ox-LDL可能通过提高Smurf2的表达抑制Ⅰ型胶原的表达;ox-LDL呈浓度依赖性逆转AngⅡ对Smurf2的抑制作用,同时抑制Ⅰ型胶原的表达。Aim To investigate angiotensin II (Ang II ) and oxidized low density lipoprotein (ox-LDL) regulating the expression of Smurt2 and collagen I in cultured rat vascular smooth muscle cells (VSMC). Methods The mRNA expression was tested by RT-PCR. The protein expression was examined by Western blot. The protein expression of collagen I was examined by ELISA. Results Compared with control group, Ang II decreased Smurf2 expression (P 〈 0. 05) and enhanced collagen I expression ( P 〈 0. 05 ), ox-LDL enhanced Smurf2 expression ( P 〈 0.01 ) and de- creased collagen I expression (P 〈 0. 05). Ox-LDL concentration-dependently induced Ang 1I -decreased Smurf2 expression (P 〈 0. 05) and collagen I concentration. Conclusion Ang II decreased Smurt2 expression in cultured VSMC. Ang II may inhibit Smurf2 expression to improve collagen I production, ox-LDL enhanced Smurf2 expression, ox-LDL may improve the expression of SmurI2 to suppress the production of collagen I. ox-LDL concentration-dependently induced Ang II-decreased Smurf2 expression, while decreased collagen I expression.
