摘 要:目的:建立总抗氧化能力评估法FRAP法微量模型。方法:以槲皮素为研究对象,通过单因素考察,建立L9(34)正交实验,对正交实验进行直观分析和方差分析。结果:由正交实验结果得出最佳实验条件为:1倍量的TPTZ溶液(0.3mol/L醋酸钠缓冲液,10mmol/LTPTZ溶液,20mmol/L氯化铁溶液三者按照顺序体积比为10∶1∶1),加入量为200μL,在37℃下反应时间为40min。验证实验结果RSD值为0.082%,表明该方法稳定可靠,本实验以PG、BHA、BHT做阳性对照,结果表明,此模型准确可行。结论:实验结果表明,本次实验建立的FRAP总抗氧化能力评估法准确,用药量小,稳定可行。Objective:To found micro-model for FRAP assay. Methods:The orthogonal experiment was designed according to L9 (34) orthogonal table after determining the single factor exploration using quercetin as antioxidant. Results: By the direct-viewing analysis and variance analysis of the results of the orthogonal experiment, the optimum experiment conditions were confirmed that the amount of TPTZ solution was i time(Sodium acetate buffer was 0.3 mol/L,TPTZ solution was 10mmol/L, and ferric chloride solution was 20mmol/L.The order volume ratio was 10:1:1 ).The addition volume of FRAP was 200μL and 40min was kept in 37℃. RSD of the evaluation test was 0.082%,and it showed the method was stable.PG, BHA and BHT were used as standard control,the model was further certified that it was precision and feasibility.Conclusion:The results indicated that the micro-model for FRAP assay from quercetin was accurate,little sample and reliable.
