摘 要:通过对聚乙二醇(PEG)-K2HPO4、PEG-(NH4)2SO4、PEG-C6H5O7Na3三种双水相体系进行研究,制作相图,分析得出PEG-K2HPO4是萃取蛋清溶菌酶较为合适的双水相体系。选用分子量适中的PEG2000与K2HPO4组成双水相体系分离萃取蛋清溶菌酶。研究表明,采用PEG-K2HPO4双水蛋相分离蛋清溶菌酶能达到较高的分配系数和回收率,当萃取体系组成为25%(w/w)PEG2000-35%(w/w)K2HPO4时,蛋清溶菌酶的分配系数为416.94,双水相萃取过程回收率为99.47%。Polyethylene glycol (PEG)-K2HPO4, PEG-(NH4) 2SO4 and PEG-C6HsOTNa3 aqueous two-phase systems were studied in lysozyme extraction from egg white. The results indicated that PEG-K2HP04 aqueous two-phase system was best. Then the lysozyme was extracted and purified by PEG2000-K2HPO4 aqueous two-phase system,which exhibited an efficient distribution coefficient and reeovery. While,the separation system composed of 25% (w/w)PEG2000-35% (w/w) K2HPO4 was found to be efficient with a distribution coefficient of 416.94,a recovery of 99.47%.
