摘 要:目的以牛至药材为研究对象寻找更简单、准确、快速、重现性好、样品用量少的中药材挥发性成分提取与分析联用分析方法。方法采用固相微萃取技术(SPME),分别用100μm PDMS、85μm PA、65μm CW/DVB、75μm Carboxen/PDMS四种类型萃取纤维头作为考察条件,结合GC-MS对湖北罗田产牛至药材中挥发性成分进行了分析研究;并与传统方法水蒸气蒸馏提取挥发油样品进行比较研究,优选牛至挥发性成分分析的最佳条件,对不同产地牛至药材中挥发性成分进行了分析研究。结果确定最佳萃取分析条件为:牛至药材取样量0.5 g,在80℃预热30 min,再采用85μm PA型Fiber,SPME萃取20 min。结论 SPME分析结果表明,不同产地的牛至挥发性成分其成分种类和相对含量上均有一定的差异,但都含有3-辛酮等8种主要成分,相对含量均超过70%。Objective Origanum vulgare L. is the object,in order to find one technique that was very easy for applying, fast, ac- curate and repeatable. Further more, it needs very little amount of sample and solvent - free. This will enable us to control the qual- ity of Chinese herbal medicines with reliable and convenient techniques. Methods Headspace solid - phase micro - extraction ( HS - SPME) combined with GC - MS technology was applied to detect the volatiles of the aerial part of the Origanum vulgare L. plant species collected from different areas in China. A preliminary screening of SPME fibers of various coatings was carried out and the extracting results were all compared with Steam Distillation Extraction (SDE) sample in order to choose the best type for the extraction of the volatiles in Origanum vulgate L. Results The best analysis condition is:85 μm PA fiber,0.5g plant material, preheating for 30min and SPME for 20min after that at 80℃. Conclusion All the experiments carried out on Origanum vulgare L. samples from different areas in China were with optimized conditions. Experiment results showed some differences within the area variation, but all those samples contained the same eight compounds, for example, 3 - octanone, further more, the relative amount of these 8 compounds in all samples were above 70%.
