摘　　要：目的　探讨特异性反义寡脱氧核苷酸 (antisenseoligodeoxynucleotide ,ASODN)对人胃癌细胞端粒酶活性的影响。方法　应用脂质体包裹与端粒酶RNA模板区互补的ASODN ,处理人胃癌细胞系FGC85。用端粒重复扩增分析 (telomererepeatamplificationprotocol,TRAP) PCR ELISA检测法观察端粒酶活性的变化。结果　经脂质体包裹ASODN处理的胃癌细胞端粒酶活性受到明显抑制 ,并与ASODN的浓度和处理时间相关。结论　端粒酶RNA模板区ASODN能够抑制端粒酶的活性 。Objective To investigate the effect of specific antisense oligodeoxynucleotide (ODN) on telomerase activity of human gastric cancer cell line FGC 85 . Methods FGC 85 cell were treated with ODN complementary to the template region of RNA component of human telomerase in vivo. A cationic liposomal transfection reagent was used as delivery vector. The changing of telomerase activity was assayed by TRAP PCR ELISA. Results Telomerase activity was suppressed significantly by antisense ODN and the effect was related with concentrations of antisense ODN and treating time. Conclusions Specific antisense ODN inhibits telomerase activity of human gastric cancer cell line in vivo, it could be used in the research of cancer gene therapy and the exploration of relationship between telomerase and neoplasm.
