摘 要:目的探讨RhoC、PRL-3及FMNL2在结直肠癌(CRC)中的表达及其相互作用关系。方法利用免疫组化、Western blot检测在不同结直肠癌细胞中RhoC、PRL-3蛋白含量变化;用Realtime-RTPCR法分析RhoC、PRL-3mRNA表达情况。结果免疫组化显示FMNL2基因沉默后,RhoC的平均光密度值(AOD)明显下降,而PRL-3的AOD值未见明显差异。随着结直肠癌细胞中FMNL2蛋白水平下调,RhoC蛋白水平表达明显下降,RhoC mRNA的表达水平是SW620细胞的0.618倍,差异有统计学意义(P<0.05),而PRL-3蛋白含量及mRNA的表达水平均未见明显差异。结论 FMNL2可能参与RhoC调控转移机制,而FMNL2对PRL-3的表达可能无反馈性调节作用。Objective The aim of this study was to investigate the expression of RhoC and PRL-3 proteins in FMNL2 silenced colorectal carcinoma (CRC). Methods The protein expression of RhoC, PRL-3 and FMNL2 was detected by immunohistochemistry in different human colorectal cancer cell lines. The mRNA and protein expression was detected by real- time RT-PCR and Western blot, respectively. Results Results from immunohistochemical staining showed that the expression of RhoC was significantly decreased in FMNL2 silenced SW620 cell lines. However, the expression of PRL-3 was not changed. Conclusion The expression of RhoC might be regulated by FMNL2 and RhoC might promote the distant metastasis of cancer cell by FMNL2.
