摘  要:目的研究诱导型一氧化氮合酶(iNOS)基因rs2297518、rs8081248位点多态性与脑动脉粥样硬化(CAA)及颅内动脉粥样硬化(ICA)的关系。方法收集患者324例,根据脑血管狭窄部位与程度将其分为CAA组243例和对照组81例,CCA组又分ICA患者137例为ICA亚组。分析各组患者脑卒中传统危险因素,并比较rs2297518、rs8081248位点的基因型频率及等位基因频率在各组的分布。结果 CAA组及ICA亚组的脑卒中传统危险因素明显高于对照组。CAA组中rs2297518位点的基因型GG及等位基因G频率分布均明显高于对照组(P<0.05);ICA亚组中rs2297518位点的等位基因G频率分布明显高于对照组(P<0.05)。结论 iNOS基因rs2297518位点的多态性与CAA及ICA有关,基因型GG可能是CAA的易感因素,等位基因G可能是CAA及ICA的易感因素;iNOS基因rs8081248位点的多态性可能与CAA或ICA无关。Objective To study the correlation of iNOS polymorphism at rs2297518 and rs8081248 with CAA and ICA. Methods Clinical data and blood samples were collected from 324 patients who underwent cerebral angiography. The patients were divided into CAA group(n=243) and control group(n=81) according to the site and severity of their cerebral artery stenosis. One hundred and thirty seven patients in CAA group served as an ICA subgroup. Traditional risk factors for cerebral infarction in different groups were analyzed. The distributions of genotype and allele frequencies at rs2297518 and rs8081248 in different groups were compared. Results The preva- lence of traditional risk factors for cerebral infarction was significantly higher in CAA group and ICA subgroup than in control group. The distribution rate of GG genotype and G allele frequencies at rs2297518 was significantly higher in CAA group than in control group(P〈0.05). No significant difference was found in GG genotype and G allele frequencies at rs8081248 between CAA group and control group. The distribution rate of G allele frequencies at rs2297518 was significantly higher in ICA subgroup than in control group(P〈0.05). Conclusion Polymorphism of iNOS gene at rs2297518 is related with CAA and ICA. GG genotype may be a susceptible factor for CAA whereas G allele may be a susceptible factor for CAA and ICA. Polymorphism of iNOS gene at rs8081248 is not related with CAA and ICA.
